Dosage of Crizotinib
The standard dose of Crizotinib is 250mg orally twice daily and should be taken with meals to increase bioavailability. For special populations, the dose should be adjusted based on liver function or drug interactions.
1. Standard usage and dosage
(1) Adult patients: The recommended dose is 250mg orally, twice a day, and continued until disease progression or intolerable toxicity occurs.
(2) Usage: The tablet should be swallowed whole and should not be crushed or chewed. It is recommended to take it with food to increase absorption rate (after meals, the bioavailability is increased by 43%).
2 dose adjustment plan
(1) Liver dysfunction: Moderate damage (Child Pugh B grade) should be reduced to 200mg twice daily, and severe damage (Child Pugh C grade) should be reduced to 250mg once daily.
(2) Drug interactions: When combined with strong CYP3A inhibitors, the dosage should be reduced by 50%, and when combined with strong CYP3A inducers, the dosage should be increased to 300mg twice daily.
3 Adverse Reaction Management
(1) Grade 3 non hematological toxicity: Suspend administration until it returns to Grade 1, and then restart treatment twice daily at 200mg.
(2) Grade 4 hematological toxicity: permanent discontinuation of medication is required, or treatment can be resumed with a reduced dose of 150mg twice daily after evaluation.
