Guidelines for taking Ceritinib: Fasting requirements and dosage management

To ensure the efficacy of Ceritinib and reduce the risk of adverse reactions, please strictly follow the following rules for administration and adjustment.

1、 Core Medication Requirements: Fasting

Time limit: At least 1 hour before taking the medication and 2 hours after taking the medication, it is forbidden to eat, drink calorie containing beverages (such as juice and milk) or nutritional supplements.

Operation suggestion: It is recommended to take the medicine at the same time every day (such as 8am), swallow the whole pill with water, and do not crush or chew it.

2、 Dose adjustment principle (based on adverse reactions)

Starting and reduction steps: The standard starting dose is 750mg/day (once daily). If severe intolerance occurs (such as hepatotoxicity or gastrointestinal reactions), gradually decrease by 150mg per day (i.e. 750 → 600 → 450 → 300mg/day).

Recovery and discontinuation: After symptom relief to ≤ level 1, it can be restored to the previous tolerated dose. If unable to tolerate 300mg/day, permanent discontinuation is required.

3、 Leakage Handling Plan

Supplement window: If missed and remembered within 12 hours, the current dose should be immediately supplemented.

Skip rule: If it exceeds 12 hours or is close to the next dose, skip the missed dose and take the next dose according to the original plan. Double dose supplementation is strictly prohibited.

Post vomiting treatment: If vomiting occurs after taking medication, do not take it again and wait for the next regular medication time.

4、 Important Interaction Reminder

During medication, avoid using potent CYP3A4 inhibitors (such as clarithromycin) or inducers (such as rifampicin) in combination, and adjust concomitant medications after evaluation by a doctor.
