Erdafitinib: Usage, Dosage, Efficacy, and Precautions
Edatinib is an oral FGFR kinase inhibitor used to treat locally advanced or metastatic urothelial carcinoma carrying FGFR2/3 gene mutations. Accurately mastering its usage and risks is the key to ensuring efficacy and safety.
1、 Usage and dosage
Initial dose: 8 milligrams once daily.
Dose escalation: Between days 14 and 21, if the blood phosphorus level is below 5.5mg/dL and there is no significant eye or skin toxicity, the dose will be increased to once daily, 9 milligrams per dose.
Dosage: Take the whole tablet on an empty stomach (at least 1 hour before or 2 hours after meals), do not crush or chew.
2、 Efficacy performance
The therapeutic effect is mainly reflected in tumor shrinkage and delaying disease progression.
Before treatment, genetic testing is required to confirm the presence of FGFR2/3 mutations.
Usually used for patients with disease progression during or after platinum based chemotherapy.
3、 Core precautions
Eye toxicity: May cause central serous retinopathy or retinal pigment epithelium detachment. Regular ophthalmic examinations should be performed before and during treatment, and immediate medical attention should be sought if symptoms such as visual blurring occur.
Hyperphosphatemia: Almost all patients will experience elevated blood phosphorus levels, and regular monitoring of blood phosphorus is necessary to adjust dosage or suspend medication based on blood phosphorus levels.
Skin and nail reactions: Hand foot syndrome, paronychia, etc. may occur, and skin care needs to be strengthened. In severe cases, treatment should be interrupted.
Oral inflammation: Pay attention to oral hygiene, and promptly treat painful ulcers or redness.
Embryo fetal toxicity: Women of childbearing age should strictly use contraception during treatment and one month after discontinuation of medication.
