The administration of Futibatinib should strictly follow the principle of fasting, and the dosage should be adjusted according to adverse reactions and special population conditions. The following provides detailed guidelines on medication methods, dosage adjustment standards, and discontinuation indications.

1. Medication method

Fubatinib should be taken on an empty stomach, and it is recommended to administer it 1 hour before or 2 hours after meals. Avoid taking with high-fat diet to prevent affecting drug absorption. Taking medication at a fixed time every day can maintain stable blood drug concentration.

2. Dose adjustment standards

(1) Liver dysfunction: Children Pugh A/B grade patients need to reduce their dosage to 80mg/day, while C grade patients are contraindicated.

(2) Renal insufficiency: eGFR<30mL/min is prohibited, and 30-89mL/min does not require adjustment.

(3) Adverse reactions: Grade 3 non hematological toxicity needs to be temporarily suspended until reduced after recovery, and Grade 4 considers permanent discontinuation of medication.

3. Indications for discontinuation of medication

(1) Irreversible organ damage: such as liver failure and severe corneal lesions.

(2) Grade 4 hematological toxicity: Neutrophils<0.5 × 10 ^ 9/L for more than 7 days.

(3) Repeated grade 3 toxicity: recurrence even after reducing the same adverse reaction.

4. Special precautions

When used in combination with strong CYP3A inhibitors, the dosage should be reduced by 50mg, and when used in combination with inducers, the dosage should be increased. Regular monitoring of liver and kidney function as well as eye condition is required during the treatment period.
