Pacritinib has shown significant efficacy in the treatment of hematological malignancies such as myelofibrosis, especially in patients with thrombocytopenia. This drug targets the JAK2/FLT3 pathway through a unique mechanism of action, with high selectivity and relatively few side effects.

1. Clinical efficacy data

In phase III clinical trials, treatment with pakinib showed significant reduction in spleen volume and improvement in symptoms in patients with myelofibrosis. About 30% of patients have a spleen volume reduction of ≥ 35%, and over 40% of patients have an overall symptom score improvement of ≥ 50%.

2. Special population effects

(1) Patients with thrombocytopenia

For patients with bone marrow fibrosis and platelet count<50 × 10 ^ 9/L, pazotinib shows superior efficacy compared to other JAK inhibitors without exacerbating thrombocytopenia symptoms.

(2) JAK2V617F mutation patients

The therapeutic effect is particularly significant for patients carrying JAK2V617F mutation, which can significantly reduce the allele burden of the mutation, and some patients achieve molecular remission.

3. Duration of therapeutic effect

Clinical data shows that the median duration of efficacy of pakinib is over 14 months. Some patients can achieve long-term disease control, and with prolonged treatment time, the degree of symptom improvement may further increase.

4. Compared with other drugs

Compared to similar JAK inhibitors, Parkitinib has shown better safety and tolerability in patients with thrombocytopenia, while maintaining a comparable level of efficacy. Its unique mechanism of action makes it the preferred treatment option for such patients.

5. Efficacy evaluation criteria

The efficacy of Parkitinib is mainly evaluated through indicators such as changes in spleen volume, improvement in symptom scores, stable hemoglobin levels, and reduced degree of bone marrow fibrosis. The first comprehensive efficacy evaluation can be conducted after 3-6 months of treatment.
