Quizartinib is an FLT3 tyrosine kinase inhibitor mainly used to treat specific types of acute myeloid leukemia. Its standard dose needs to be adjusted according to the patient's condition, and attention should be paid to the safety of medication for special populations.

1. Indications

Quetzatinib is indicated for the treatment of relapsed or refractory acute myeloid leukemia (AML) adult patients with FLT3-ITD mutations. This drug selectively inhibits FLT3 receptor tyrosine kinase, blocks the tumor cell proliferation signaling pathway, and is suitable for targeted therapy after standard treatment failure.

2. Usage and dosage

(1) Standard dose

The recommended dosage for adult patients is once daily oral administration of 30mg, which should be taken on an empty stomach (1 hour before or 2 hours after meals). Treatment should continue until disease progression or intolerable toxic reactions occur.

(2) Dose adjustment

When QT interval prolongation occurs, administration should be suspended until recovery, and subsequently reduced to 20mg per day. If other grade 3 or above adverse reactions occur, administration should be suspended until recovery to ≤ grade 1, and treatment should be continued at the same or reduced dose.

3. Medication for special populations

(1) Liver dysfunction

Mild liver dysfunction does not require dose adjustment, and patients with moderate to severe liver dysfunction should use it with caution. It is recommended to start with lower doses and closely monitor.

(2) Renal insufficiency

Mild to moderate renal impairment does not require dose adjustment, and patients with severe renal impairment have limited data and should be used with caution under the guidance of a doctor.

(3) Elderly patients

Patients aged 65 and above do not need to adjust the starting dose, but due to their potential sensitivity to medication, it is necessary to strengthen adverse reaction monitoring.

(4) Pregnancy and lactation

Having embryonic fetal toxicity, women of childbearing age need to take effective contraceptive measures. Breastfeeding women should stop breastfeeding or medication.
