Correct usage of elacestrant
The standard usage of elacestrant is once daily oral administration of 345mg, which should be taken as a whole tablet until disease progression or intolerable toxicity occurs. Attention should be paid to the handling of missed doses, dosage adjustments, and medication contraindications for special populations, especially the risk of blood lipid abnormalities and embryonic toxicity.
1. Conventional usage and dosage
(1) Basic dose: Take 345mg orally once a day (four tablets of 86mg each), unaffected by food intake.
(2) Treatment requirements: Continue medication until disease progression or unacceptable toxic reactions occur.
2 Omission and Overtreatment
(1) Missed dose: If the medication has not been taken for more than 6 hours, the dose should be skipped and taken according to the original plan the next day; If vomiting occurs after taking medication, it is necessary to take additional medication.
(2) Overdose: Seek medical attention immediately and receive supportive treatment based on symptoms.
3 dose adjustment plan
(1) First dose reduction: 258mg (three tablets of 86mg), suitable for moderate liver damage or specific adverse reactions.
(2) Secondary dose reduction: 172mg (two tablets of 86mg), used for recurrent grade 3/4 toxicity.
(3) Permanent discontinuation of treatment: Discontinue treatment when grade 4 toxicity recurs or cannot be tolerated.
4. Medication for special populations
(1) Patients with liver damage: Moderate to severe liver dysfunction should be reduced or disabled, and mild cases should not be adjusted.
(2) Pregnant/breastfeeding period: Contraception is prohibited and strict measures must be taken. Breastfeeding is prohibited within one week after discontinuing medication during breastfeeding.
(3) Maternity protection: Efficient contraceptive measures should be taken during treatment and one week after discontinuation of medication.
5 Key Precautions
(1) Blood lipid monitoring: 30% of patients have abnormal blood lipids and need regular testing for cholesterol and triglycerides.
(2) Tablet administration: The entire tablet should be swallowed and should not be broken or chewed.
