Daraxonrasib is an inhibitor of the RAS GTPase family, indicated for the treatment of adult patients with metastatic pancreatic adenocarcinoma who have received at least one prior systemic therapy or who are not candidates for multiagent systemic therapy.

Dosage and Administration:

The recommended dosage is 300 mg orally once daily, with or without food. Swallow tablets whole; do not chew, crush, or split.

Take at the same time each day. If a dose is missed for more than 4 hours, skip the missed dose and take the next dose at the next regularly scheduled time; if vomiting occurs after taking the dose, do not take an additional dose.

Treatment should continue until disease progression or unacceptable toxicity.

Before and during treatment, topical corticosteroids and emollient creams are recommended, sun exposure should be limited, sunscreen should be used, and prophylactic oral antibiotics may be considered to reduce the risk of skin reactions.

Efficacy:

In the RASolute 302 study, daraxonrasib demonstrated statistically significant improvements in overall survival, progression-free survival, and objective response rate in both the RAS G12 mutation population and the overall population, with efficacy superior to physician's choice of standard chemotherapy regimens.

Precautions:

Common adverse reactions include rash, diarrhea, stomatitis, nausea, fatigue, vomiting, abdominal pain, edema, decreased appetite, and hemorrhage.

Monitor for dermatologic and soft tissue toxicity, stomatitis, diarrhea, gastrointestinal perforation, and interstitial lung disease/pneumonitis; withhold, reduce the dose, or permanently discontinue based on severity.

This product can cause fetal harm. Females and males of reproductive potential should use effective contraception during treatment and for 1 week after the last dose. Women should not breastfeed.

Avoid concomitant use with strong CYP3A inhibitors with P-gp inhibition and cyclosporine A; dose adjustments are required when used with strong or moderate CYP3A inhibitors or inducers; P-gp substrate drugs should be taken at least 4 hours apart from this product.

Patients should inform their doctor of all concomitant medications and undergo regular monitoring of relevant parameters.
