Sutoraxib (trade name Lumakras/AMG510) is a targeted drug used for specific cancers, with a standard dose of 960mg orally per day. Attention should be paid to risks such as liver toxicity and interstitial lung disease, and medication plans should be adjusted for specific populations. 1. Usage and dosage specifications

(1) Standard dose: 960mg orally once daily for non-small cell lung cancer/colorectal cancer patients, taken at a fixed time to maintain stable blood drug concentration.

(2) Dose adjustment: In case of grade 3-4 adverse reactions, medication should be suspended and gradually reduced from 480mg to 240mg after recovery. For interstitial lung disease, medication should be permanently discontinued.

(3) Medication management: Medication can be taken within 6 hours of missed intake, but not after vomiting; Avoid using strong CYP3A inducers in combination to affect drug efficacy.

Analysis of Drug Resistance Mechanisms

(1) Target mutation: KRASG12C mutation may result in secondary mutations (such as Y96D, R68S), leading to conformational changes in the drug binding site.
