Talazoparib, as a PARP inhibitor, has shown significant efficacy in specific cancer treatments, especially for tumor patients carrying BRCA gene mutations. It exerts anti-tumor effects by interfering with the DNA repair mechanism of tumor cells, and clinical data shows that it can significantly prolong progression free survival.

1. Main indications

(1) BRCA mutation in advanced breast cancer: based on EMBRACA study, talazopari significantly prolonged the progression free survival period (8.6 months vs 5.6 months) compared with chemotherapy;

(2) Maintenance therapy for ovarian cancer: SOLO series studies confirm its survival benefits for BRCA mutation patients;

(3) Prostate cancer: TRITON2 study shows effectiveness for patients with DNA repair defects.

2. Clinical efficacy data

(1) Objective remission rate: 62.6% of patients with breast cancer and 50-60% of patients with ovarian cancer;

(2) Median duration of remission: 5.4 months for breast cancer patients and 11.2 months for ovarian cancer patients;

(3) Disease control rate: Overall, it can reach over 80%.

3. Advantages of mechanism of action

(1) Strong PARP capture ability: inhibits PARP enzyme activity while trapping it at DNA damage sites;

(2) Dual mechanism of action: blocking DNA single strand break repair and causing double strand breaks;

(3) High selectivity: The inhibitory activity against PARP-1/2 is significantly higher than other PARP subtypes.

4. Special population effects

(1) BRCA mutation patients benefit the most significantly;

(2) HRD positive patients are second;

(3) The effect is better for platinum sensitive patients.

5. Time window for missed medication supplementation

If you find that you have missed taking Tarazopanib, it is recommended to take it as soon as possible within 12 hours. This is consistent with the treatment principles of PARP inhibitors such as razetinib. If it exceeds 12 hours, it is not recommended to take the next dose according to the original plan.

6. Risk of drug overdose

At present, there is no specific antidote for overdose of Terazopanib. Based on the experience of similar drug treatments, close monitoring and targeted supportive treatment are necessary.
