What are the contraindications for Abemaciclib in Laos and its use in special populations
The contraindications for Abemaciclib mainly involve allergic reactions, pregnancy, and severe liver injury. For special populations, the dosage should be adjusted or protective measures taken according to liver and kidney function, age, and fertility status.
1. Contraindications
(1) It is contraindicated for individuals who are allergic to Abexili or its excipients, as severe hypersensitivity reactions such as angioedema or anaphylactic shock may occur.
(2) Pregnant women are prohibited from using it, animal studies have shown embryotoxicity (such as skeletal deformities), and human data is limited but the risks are clear.
(3) It is contraindicated for patients with Child Pugh C grade liver dysfunction, as significant inhibition of drug metabolism may lead to toxic accumulation.
2. Medication for special populations
(1) Pregnancy and lactation period
Women of childbearing age need to confirm their pregnancy status before treatment, and effective contraception is required during medication and 3 weeks after the last dose; Prohibited during lactation to avoid drugs affecting infants through milk.
(2) Liver dysfunction
Child Pugh A/B patients should reduce their dosage to 150mg or 100mg twice a day and closely monitor their transaminase and bilirubin levels.
(3) Renal insufficiency
Mild to moderate damage (CLcr ≥ 30mL/min) does not require dose adjustment; Severe damage or insufficient data on dialysis patients, it is recommended to use with caution and monitor toxicity.
