Capmatinib is a MET inhibitor used to treat metastatic non-small cell lung cancer with MET exon 14 jump mutation. The dosage should be adjusted according to the patient's liver function, and caution should be exercised in assessing the risks and benefits of medication for special populations.

1. Indications

(1) Suitable for treating adult patients with metastatic non-small cell lung cancer carrying MET exon 14 skipping mutation.

(2) The MET mutation status needs to be confirmed through FDA approved testing methods.

2. Usage and dosage

(1) The recommended dosage is 400mg twice daily, taken orally and can be taken with food or on an empty stomach.

(2) Patients with severe liver dysfunction should reduce their dosage to 200mg twice a day.

(3) If serious adverse reactions occur, medication can be suspended or reduced to twice daily, 200mg each time.

3. Medication for special populations

(1) Pregnancy: May cause harm to the fetus, pregnant women and women with fertility potential need to use effective contraceptive measures.

(2) Breastfeeding period: It is recommended that lactating women avoid breastfeeding during the medication period and within one week after discontinuing the medication.

(3) Elderly patients: Patients aged 65 and above do not need to adjust the dosage, but adverse reactions need to be closely monitored.

(4) Liver dysfunction: Patients with mild to moderate liver dysfunction do not need to adjust the dosage, while patients with severe liver dysfunction need to reduce the dosage.

4. Precautions

(1) Regular monitoring of liver function, especially ALT and AST levels, is necessary during treatment.

(2) Interstitial lung disease may occur. If there is difficulty breathing or worsening cough, medication should be stopped immediately and medical attention sought.

(3) Avoid co administration with potent CYP3A4 inducers or inhibitors as it may affect drug concentration.
